چکیده

Contraction in skeletal muscle is initiated when Ca stored in the SR is released into the myoplasm in response to membrane depolarization. Studies of the mechanisms that control SR Ca release are aided by methods that permit estimating the Ca permeability of the SR membrane or, equivalently, the extent of activation of the SR Ca release channels. This quantity is proportional to the rate of Ca release divided by the driving force for release. Because the free calcium concentration in the myoplasm ([Ca]myo) is generally small relative to that in the SR ([Ca]SR), and because the SR membrane potential is expected to be near zero (Somlyo, et al., 1977; Labarca and Miller, 1981; Garcia and Miller, 1984; Pape et al., 1992; Gillespie and Fill, 2008), the driving force for Ca release should be proportional to [Ca]SR.

Calcium buffering properties of sarcoplasmic reticulum and calcium-induced Ca2+ release during the quasi-steady level of release in twitch fibers from frog skeletal muscle

Experiments were performed to characterize the properties of the intrinsic Ca(2+) buffers in the sarcoplasmic reticulum (SR) of cut fibers from frog twitch muscle. The concentrations of total and free calcium ions within the SR ([Ca(T)](SR) and [Ca(2+)](SR)) were measured, respectively, with the EGTA/phenol red method and tetramethylmurexide (a low affinity Ca(2+) indicator). Results indicate S...

Major contribution of sarcoplasmic reticulum Ca depletion during long-lasting activation of skeletal muscle

Depolarization of skeletal muscle fiber induces the release of Ca ions from the SR through RyR Ca release channels, which in turn activate contraction. Activity of the RyR is a tightly voltage-controlled process that involves the dihydropyridine receptors (DHPRs) anchored in the t-tubules, which act as voltage sensors and transduce membrane potential change into RyR opening (Ríos and Pizarro, 1...

Effects of fatigue on sarcoplasmic reticulum and myofibrillar properties of rat single muscle fibers.

Force decline during fatigue in skeletal muscle is attributed mainly to progressive alterations of the intracellular milieu. Metabolite changes and the decline in free myoplasmic calcium influence the activation and contractile processes. This study was aimed at evaluating whether fatigue also causes persistent modifications of key myofibrillar and sarcoplasmic reticulum (SR) proteins that cont...

Calcium dependence of inactivation of calcium release from the sarcoplasmic reticulum in skeletal muscle fibers

The steady-state calcium dependence of inactivation of calcium release from the sarcoplasmic reticulum was studied in voltage-clamped, cut segments of frog skeletal muscle fibers containing two calcium indicators, fura-2 and anti-pyrylazo III (AP III). Fura-2 fluorescence was used to monitor resting calcium and relatively small calcium transients during small depolarizations. AP III absorbance ...

Methyl Jasmonate-Induced Stimulation of Sarcoplasmic Reticulum Ca -ATPase Affects Contractile Responses in Rat Slow-Twitch Skeletal Muscle

The purpose of this study was to determine whether methyl jasmonate, a stimulator of Ca -adenosine triphosphatase (ATPase) activity of the purified ATPase from fast-twitch skeletal muscle, could affect contractile responses in small bundles of rat isolated slow-twitch (soleus) fibers. In saponin-skinned fibers, sarcoplasmic reticulum (SR) Ca loading was performed in pCa 7.0 solution. The amount...
